Kappa mRNA CISH
Immunohistochemistry Stain
Useful For
In situ mRNA detection of Kappa and Lambda light chain expression is an important tool used in detection of clonal population of infiltrating plasma cells in bone marrow, expressing either Kappa or Lambda immunoglobulin light chain cytoplasmic mRNA, may serve as a valuable indicator of disease states. Kappa and Lambda mRNA CISH is intended for the detection of Kappa and Lambda immunoglobulin light chain mRNA.
Method name and description
- Kappa mRNA CISH
- Chromogenic In-Situ Hybridization (CISH) stain on formalin-fixed, paraffin-embedded (FFPE) tissue section
Clinical information
Kappa and Lambda mRNA CISH are a set of oligonucleotide-based probes that bind to either the Kappa or Lambda immunoglobulin light chain mRNA. The probes are conjugated with fluorescein isothiocyanate (FITC) to enable detection using the Anti-FITC-AP CISH.

Specimen type / Specimen volume / Specimen container
- Specimen type: Well-fixed tissue in 10% neutral buffered formalin.
- Submit 2 unstained (3-4 µm thick) paraffin embedded tissue section mounted on a clean positively charged glass slide. One slide will be stained for ISH Lambda and the other slide will be stained for the ISH +ve probe (the use of the ISH +ve probe is for evaluation of RNA quality by chromogenic in situ hybridization (CISH)) (sections to be cut within 6 weeks.) or formalin-fixed, paraffin-embedded (FFPE) tissue block
Storage and transport instructions
- Slides or blocks are stored in cork box at room temperature away from sun light and any source of heat.
- Follow your local regulation shipping guidelines.
Specimen Rejection Criteria
- Broken slides
- Unlabeled slides with patient/case identification
- Contaminated slides
- Slides/paraffin blocks mismatch
- Uncharged slides
Factors affecting test performance and result interpretation
- Fixation time (FT)
- Fixative Type
- Storage time in paraffin
- Storage temperature
- Age of the cut sections
- Section thickness
Turnaround time / Days and times test performed / Specimen retention time
- Turnaround time for the test in platform: 4-5 hours
- Days and times test performed: Once a week (0600H- 1400H)
- Note: Request received after the staining day will be stained the following week.
- Shelf-life of the paraffin section slides: ≤6 weeks
