Qualitative measurement of the circulating IgE antibodies to a mix of specific grass pollens allergen gx2 (Bermuda grass, Rye-grass, timothy grass, Meadow grass, Johnson grass, Bahia grass) in human serum help to identify the trigger that may be contributing to the symptoms and confirm sensitization to a particular allergen. 

Method name and description

Method: Fluorescence Enzyme Immunoassay (FEIA), Uses the ImmunoCAP method on Phadia system.

The ImmunoCAP solid phase consists of a cellulose derivative specific for allergen enclosed in a capsule. The FEIA technology is based on an extremely high total binding capacity this ensures binding of all relevant antibodies.

The allergen of interest, covalently coupled to ImmunoCAP, reacts with the specific IgE in the patient sample. After washing away non-specific IgE, enzyme labeled antibodies against IgE are added to form a complex. Following incubation, unbound enzyme-anti IgE is washed away and the bound complex is then incubated with a developing agent. After stopping the reaction, the fluorescence of the eluate is measured. The higher the response value, the more specific IgE is present in the sample.

Clinical information

Clinical manifestations of immediate hypersensitivity (allergic) diseases are caused by the release of proinflammatory mediators (histamine, leukotrienes, and prostaglandins) from immunoglobulin E (IgE)-sensitized effector cells (mast cells and basophils) when cell-bound IgE antibodies interact with allergen.

In vitro serum testing for IgE antibodies provides an indication of the immune response to allergen(s) that may be associated with allergic disease.

IgE - Immunoglobulin E - is a type of protein called an antibody. It plays an important role in allergy reactions and is therefore often called the “allergy antibody”.

When the person is exposed to a particular substance (allergen), the immune system starts the production of IgE as an attempt to protect the body. The specific IgE antibodies remain in the body and next time will react with specific allergen on subsequent exposure.

Storage and transport instructions

For sample transportation, the sample should remain at room temperature (18-25°C) for no longer than eight hours.

Separated serum samples can be stored at 2-8°C for one week.

Separated serum samples should be frozen at below -20°C for any long-term storage.

Interpretation:

*Positive results indicate the possibility of allergic disease induced by one or more allergens present in the multi-allergen grass pollens mix (gx2).

*Negative results may rule out allergy, except in rare cases of allergic disease induced by exposure to a single allergen.

Factors affecting test performance and result interpretation

Testing for IgE antibodies is not useful in patients previously treated with immunotherapy to determine if residual clinical sensitivity exists, or in patients in whom the medical management does not depend upon identification of allergen specificity.
