Test ID: IH HMB45
Melanosome
Immunohistochemistry Stain
Useful For

Identification of Melanocytes with immature melanosome formation in normal skin, nevus, and melanoma tissue. Useful in Angiomyolipoma and lymphangioleiomyoma.

Method name and description
  • Monoclonal Mouse Anti-Human Melanosome, Clone HMB45
  • Immunoperoxidase stain on formalin-fixed, paraffin-embedded (FFPE) tissue section
Reporting name

HMB45

Clinical information

The presence of the antigen indicates active melanosome formation and thus melanocytic differentiation. It is also expressed in normal fetal melanocytes, but not in normal resting adult melanocytes, regardless of the degree of pigmentation. Upon activation, adult melanocytes can re-express the HMB45-define antigen (as it is expressed in fetal melanocytes). Such melanocytes are activated by variety of stimuli. For example, HMB45-positive cells have been detected in tissue overlying or adjacent to the granulation tissue, hemangiomas, vessel-rich tumor stroma, and basal cell carcinoma. Hair follicle stain occasionally due to associated stimulated melanocytes. Positive HMB45 staining has not been observed with melanocyte in lentigines or overlying fibroblastic proliferation such as keloids, dermatofibromas, and old fibrotic hemangiomas. Non-melanocytic normal tissues do not react with HMB45 antibody.

Specimen type / Specimen volume / Specimen container
  • Specimen type: Well-fixed tissue in 10% neutral buffered formalin.
  • Submit 2 unstained (3-4 µm thick) paraffin embedded tissue section mounted on a clean positively charged glass slide  (sections to be cut within 6 weeks.) or formalin-fixed, paraffin-embedded (FFPE) tissue block.
Storage and transport instructions
  • Slides or blocks are stored in cork box at room temperature away from sun light and any source of heat.
  • Follow your local regulation shipping guidelines.
Specimen Rejection Criteria
  • Broken slides
  • Unlabeled slides with patient/case identification
  • Contaminated slides
  • Slides/paraffin blocks mismatch
  • Uncharged slides
