Clinical information

Alternative splicing gives rise to two mRNA variants from the human OCT4 gene on chromosome 6, originally named OCT3A and OCT3B, thus the adapted OCT3/4 name. The OCT3A and OCT3B isoforms are composed of 360 and 265 amino acids, respectively, of which the 225 amino acids at the C-termini are identical. OCT3/4 belongs to the POU (Pit-Oct-Unc) transcription factor family which can activate the expression of their target genes through binding an octameric sequence motif of an AGTCAAAT consensus sequence. OCT3/4 is expressed in early embryonic cells and germ cells and is central to the gene regulatory network responsible for self-renewal, pluripotency, and lineage commitment in embryonic stem cells and induced pluripotent stem cells. OCT3/4 expression is also detected in specific subtypes of germ cell tumors, such as seminomas, germinomas, dysgerminoma, embryonal carcinoma, the germ cell component of gonadoblastomas, and IGCNU.

  • Specimen type: Well-fixed tissue in 10% neutral buffered formalin.
  • Submit 2 unstained (3-4 µm thick) paraffin embedded tissue section mounted on a clean positively charged glass slide  (sections to be cut within 6 weeks.) or formalin-fixed, paraffin-embedded (FFPE) tissue block.
