Olig2

Clinical information

Olig2, a basic helix–loop–helix transcription factor, is involved in oligodendroglial specification. Olig2 expression has been reported in most glial tumors, such as oligodendrogliomas and astrocytomas. More than half of glioblastomas are weakly positive for Olig2. No Olig2 expression has been found in the non-glial tumors including neuroepithelial tumors, ependymomas, subependymomas, medulloblastomas, and non-neuroepithelial tumors, such as CNS lymphomas, meningiomas, schwannomas, atypical teratoid/rhabdoid tumor, and haemangioblastomas. In order to characterize cellular subtypes that constitute astrocytomas, oligoastrocytomas and oligodendrogliomas, double labeling of Olig2 and GFAP has been performed which identified two phenotypically distinct tumor populations. The first is Olig2+/GFAP– which has an oligodendroglial morphology, corresponding to pure oligodendrogliomas that contain only oligodendroglial cells; the second is Olig2–/GFAP+ which has an astrocytic phenotype, including not only oligoastrocytomas, but also WHO astrocytomas.

Specimen type / Specimen volume / Specimen container
  • Specimen type: Well-fixed tissue in 10% neutral buffered formalin.
  • Submit 2 unstained (3-4 µm thick) paraffin embedded tissue section mounted on a clean positively charged glass slide  (sections to be cut within 6 weeks.) or formalin-fixed, paraffin-embedded (FFPE) tissue block.

Storage and transport instructions
  • Slides or blocks are stored in cork box at room temperature away from sun light and any source of heat.
  • Follow your local regulation shipping guidelines.
Specimen Rejection Criteria
  • Broken slides
  • Unlabeled slides with patient/case identification
  • Contaminated slides
  • Slides/paraffin blocks mismatch
  • Uncharged slides
